- Klinghoffer was born in Galicia on February 17, 1905, and grew up in Vienna. He pursued his academic education at the University of Vienna, where he earned doctorates in both Law and Political Science.
- Following the rise of the Nazis, he fled to France and subsequently to Brazil in 1941.
- In 1953, he immigrated to Israel. He joined the academic staff of the Faculty of Law at the Hebrew University of Jerusalem, where he served as a full professor and, from 1961, as Dean of the Faculty. He specialized in administrative law and constitutional law.
- Klinghoffer was one of the founders of the Liberal Party and served as a Member of Knesset on its behalf during the Fifth, Sixth, and Seventh Knessets. On January 15, 1964, he submitted to the Fifth Knesset a proposal for a Basic Law: Bill of Fundamental Human Rights, 5724–1963—a document described by legal experts as a masterpiece—but the proposal was rejected due to opposition from the government.
- He advocated for the drafting of a constitution for the State of Israel and proposed the establishment of a second legislative chamber, similar to the systems found in many other countries.
- When Israeli television was established, he campaigned for it to operate as an independent public broadcaster, free from political influence.
- Klinghoffer served as legal counsel to the Ombudsman for the Defense Establishment from 1975 to 1976. He was the first president of the Association for Civil Rights in Israel (ACRI), serving from 1976 to 1982, and also served as the first president of the Association for Public Law in Israel. Additionally, he served as a director of the Jewish Colonial Trust (*Otzar Hityashvut HaYehudim*).
- In 1988, he was awarded the Zeltner Prize.
- He passed away on January 31, 1990.
